As a step to find out the primary replication sites of Hantaan virus, one of the causative agents of hemorrhagic fever with renal syndrome, Hantaan virus(76/118) was inoculated into the right lungs of 18 Sprague Dawley rats and bone marrows were
collected on day one, two three, four, five and seven after inoculation of virus.
From five unifected SD rats, bone marrows were collected and suspended in RPMI(Roswell Park Memorial Institute) 1640 medium, and then Hantaan virus suspension was added. Hantaan virus-infected bone marrow cells were collected on day one, two
three,
four
and five after inoculation of virus. From three uninfected SD rats, bone marrows were collected for normal control. Collected bone marrow cells were examined after staining by ordinary indirect fluorescent technic with monoclonal antibody to
capsid
protein of Hantaan virus
Hantaan virus antigens were found in bone marrow cells collected from SD rats inoculated with Hantaan virus from day one to seven, and also in bone marrow cells inoculated with virus from day one to five. Hantaan virus antigen was not found in
normal
control cells. Bone marrow cells could be the one of the sites of primary replication of Hantaan virus in SD rats.
